BMP-2 content in demineralized bone matrix for primary assessment of osteoinductive capacity of donor bone

Abstract: [Objective] To quantitate the BMP-2 content in demineralized bone matrix (DBM) for primary assessment of osteoinductive capacity of donor bone for bone bank. [Methods] The DBM sample was obtained after soft tissue removed, defatted, decalcified from the donor bone. The BMP-2 content in DBM was detected by Elisa method. Osteoinductivity was also evaluated by osteoinductive test in vivo in nude mice. The relationship between BMP-2 content and osteoinductive capacity were researched. [Results] The BMP-2 content of DBM made by the bones from different donors varied greatly, ranged from 5.85 ng/g to 71.83ng/g. In term of osteoinductive test in vivo, the bone formation area of the DBM implant significantly increased with the increase of BMP-2 content (P<0.05) . The bone formation area was <5% as the BMP-2 content in the DBM was below 30 ng/g,while increased to >25% as the BMP-2 content increased to more than 60 ng/g. [Conclusion] The content of BMP-2 in the DBM sample is positively related to the donor bone quality, which might be used for initially screening the donor bone by Elisa for BMP-2.
