Abstract: [Objective] To analyze the effect of huoxuejegu capsule on the expression of bFGF, TGF- beta and BMP-2 in tibial fracture of rabbits.
[Methods] A
total of 36 male rabbits underwent operation to create the tibial
fracture models, and were randomly divided into the huoxuejegu group and
the normal saline group with 18 animals in each group. On the first day
after operation, the suspension of huoxuejegu capsule or normal saline
was intragastrically given to the animals in corresponding groups. At 1,
3 and 5 weeks after operation, 6 animals in each group were randomly
selected and sacrificed to harvest the samples of fracture site.
Quantitative immunohistochemistry assay was conducted to measure the
expression of bFGF, TGF-¦Â and BMP-2 in fracture sites.
[Results] The
bFGF expression reached the peak at 3 weeks after operation in both
groups. Although no a significant difference in the bFGF OD value was
noticed at 1 week between the two groups (P>0.05) , the huoxuejegu
group got significantly higher bFGF OD value than the saline group at 3
and 5 weeks after operation (P<0.05) . In addition, the TGF¦Â OD value
significantly decreased in both groups as time went (P<0.05) . The
huoxuejegu group had significantly higher TGF¦Â OD value than the saline
group at 1 and 3 weeks after operation (P<0.05) , despite of no a
statistically significant difference between them at 5 weeks (P>0.05)
. Furthermore, the BMP-2 OD value significantly increased over time in
both groups (P<0.05) . Nevertheless no a statistically significant
difference in the BMP-2 expression between the two groups was proved at 1
week (P>0.05) , the huoxuejegu group had significantly higher BMP-2
OD value than the saline group at 3 and 5 weeks after operation
(P<0.05) .
[Conclusion] The huoxuejiegu capsule does promote the expression of bFGF, TGF-¦Â and BMP-2 in tibial fracture sites in rabbits.
